A kinetic analysis of the glycolytic rate and certain glycolytic enzymes in normal and leucemic leucocytes.

چکیده
It is generally agreed that human leucocytes have high aerobic glycolytic rates (1) and previous studies (2, 3) have shown the rates in homogenates of isolated normal leucocytes to be significantly higher than in leucemic cells. The present studies seek to explain this metabolic difference in terms of possible differences between the enzymic mechanisms of glycolysis in normal and leucemic leucocytes. Throughout the huge literature on glycolysis, it is often stated that the glycolytic rate of a tissue is determined by a single, rate-limiting step in the sequential chain of reactions although its identity is unknown. Nor is it known whether the rate-limiting step is the same in all tissues or whether it remains the same in pathological variants showing altered glycolytic rates. Some evidence has implicated the cell adenosine triphosphate-adenosine diphosphate ratio (4) as the determinant of glycolytic rate, a ratio at least partially controlled by the balance between adenosinetriphosphatase and hexokinase, but the data are inconclusive. Presumably, that ratio would influence the activity of adenylic acidlinked enzymes, one of which might then become rate-limiting. Burton (5) and Hinshelwood (6) have emphasized the possible hazards of the “master reaction” concept by pointing out that in a sequence of irreversible first order reactions

Background & Aim: Hexokinase and pyruvate kinase are two regulatory enzymes of glycolytic pathway in erythrocytes. Increasing evidence suggests that cigarette smoking which produces free radicals and oxidative stress can cause damage to body macromolecules such as proteins and enzymes. The aim of the present study was to investigate the susceptibility of key enzymes of erythrocytes glycolyt...

Introduction: Any type of exercise protocol has specific effects on the physiology of the body. Thus, according to the purpose of the training program and conditions of the subjects, a specific exercise protocol is needed to be considered. Therefore, the purpose of this study was to assess the effect of intermittent training on oxidative and glycolytic capacity in rat skeletal muscles. Method...
متن کاملControl of the glycolytic flux in Saccharomyces cerevisiae grown at low temperature: a multi-level analysis in anaerobic chemostat cultures.
Growth temperature has a profound impact on the kinetic properties of enzymes in microbial metabolic networks. Activities of glycolytic enzymes in Saccharomyces cerevisiae were up to 7.5-fold lower when assayed at 12 degrees C than at 30 degrees C. Nevertheless, the in vivo glycolytic flux in chemostat cultures (dilution rate: 0.03 h(-1)) grown at these two temperatures was essentially the same...
متن کاملThe association of glycolytic enzymes with cellular and model membranes.
This article deals with the binding of glycolytic enzymes with membranous or protein subcellular structures. The representative papers of the last three decades dealing with this matter are reviewed. The studies evidencing the binding of some glycolytic enzymes to insoluble subcellular proteins and membranous structures are presented. It is currently generally accepted that the glycolytic enzym...
متن کاملRegulation of human jejunal glycolytic enzymes by oral folic acid.
The effect of oral folic acid on jejunal glycolytic enzyme activity in five fasting obese patients and in three normal male volunteers on a constant 3000 cal diet was studied. The glycolytic enzymes, fructokinase, hexokinase, glucokinase, fructose-1-phosphate aldolase, and fructose diphosphate aldolase, and the disaccharidases, sucrase, maltase, and lactase were measured. In both the fasting pa...
